The exposed monocytic cells secreted interleukin 8 (IL-8) chemokine in a dose-dependent manner compared to the unexposed cell groups depicting a biologically significant inflammatory response. Results: Treatment of the cells with flavoring chemicals and flavored e-liquid without nicotine caused cytotoxicity dose-dependently. Cytotoxicity due to the exposure to selected e-liquids was assessed by cell viability and the IL-8 inflammatory cytokine response in the conditioned media. These DCF fluorescence data were expressed as hydrogen peroxide (H 2O 2) equivalents. Cell-free ROS produced by these commonly used flavoring chemicals were also measured using a 2′,7′dichlorofluorescein diacetate probe. Cell viability and the concentrations of the secreted inflammatory cytokine interleukin 8 (IL-8) were measured in the conditioned media.
Methods: Two monocytic cell types, MM6 and U937 were exposed to commonly used e-cigarette flavoring chemicals diacetyl, cinnamaldehyde, acetoin, pentanedione, o-vanillin, maltol and coumarin at different doses between 10 and 1,000 μM. We hypothesized that the flavoring chemicals used in e-juices/e-liquids induce an inflammatory response, cellular toxicity, and ROS production. The potential to cause oxidative stress by these flavoring chemicals was assessed by measuring the production of reactive oxygen species (ROS).

We focused our study on the immuno-toxicological and the oxidative stress effects by these e-cigarette flavoring chemicals on two types of human monocytic cell lines, Mono Mac 6 (MM6) and U937. I love this flavouring I can say it does change the recipe from dry and flavoursome to juicy and tastier you can't pick it out it helps blend the fruit and makes the mix mouth watering. I was hoping this would give it a fizzy type mouth feel, it doesn't at this % but it does help change the orange flavour a touch to make it more genuine almost like Fanta or tango. (Ecto cooler) I used to have this as a stand alone, it's a great tasting flavour but uses a hell of a lot, it's supposed to be reminiscent of an old school juice box from the mid - late 80's although I can barely remember that. I've spent nearly 2 months working on it trying different fruits for accents and different levels of sweetener to get it down as low as possible without losing that sweetness we all associate with commercial juice. In closing I feel I've remixed this quite nicely. I believe I used far less than they prob do but got the same effect by accentuating the Orange with the strawberries and ecto cooler. I love these in tandem and they are just the supporting note I needed to give it a fruity inhale. I wanted something to add to the fruitiness without taking over the Orange and add a bit of juicy flavor to the mix and these strawberries do just that.
